In the 1930’s, Bower and Hucker developed a medium for detecting bacteria in milk and other dairy products. Prickett used a glucose agar containing tryptone to study thermophilic bacteria in milk. This medium is known as Yeast Dextrose Agar. In 1948, the American Public Health Association (APHA) adopted Tryptone Glucose Extract Agar for use in testing milk and dairy products. Currently, APHA specifies Tryptone Glucose Extract Agar for the heterotrophic plate count procedure in testing bottled water. Baird Parker Agar is used for the cultivation of Staphylococcus aureus from foodstuffs according to ISO 6888, and is not intended for use in the diagnosis of disease or other conditions in humans.Originally introduced in 1962, this medium was developed by Baird-Parker to overcome the problems of recovering damaged Staphylococcus aureus from foodstuffs. This version of the medium is formulated according to ISO 6888-1:1999+A1:2003 and is in compliance with ISO 6888-2:2003+A1:2003 and ISO 6888-3:2003.Baird-Parker medium is highly selective by nature, due to the presence of potassium tellurite and lithium chloride. Tellurite inhibits most coliforms and is also reduced to telluride by S. aureus, giving the typical black colonies. Fluid Thioglycolate Medium is used for sterility testing. This medium conforms to Harmonized USP/EP/JP requirements and is not intended for use in the diagnosis of disease or other conditions in humans.Fluid Thioglycolate Medium is recommended by the Harmonized Pharmacopoeia for sterility testing and conforms to USP/EP/JP performance specification. In this formulation casein and yeast extract provide a source of nitrogen, essential vitamins and amino acids. The glucose provides a carbon source and sodium chloride maintains osmotic balance. L-Cystine and sodium Thioglycolate act as reducing agents to create an anaerobic environment and maintain a low Eh. This is aided by the low level of agar which reduces the oxygen permeability through the medium.
